Key Points
Question Does acetaminophen use during pregnancy increase children’s risk of neurodevelopmental disorders?

Findings In this population-based study, models without sibling controls identified marginally increased risks of autism and attention-deficit/hyperactivity disorder (ADHD) associated with acetaminophen use during pregnancy. However, analyses of matched full sibling pairs found no evidence of increased risk of autism (hazard ratio, 0.98), ADHD (hazard ratio, 0.98), or intellectual disability (hazard ratio, 1.01) associated with acetaminophen use.

Meaning Acetaminophen use during pregnancy was not associated with children’s risk of autism, ADHD, or intellectual disability in sibling control analyses. This suggests that associations observed in other models may have been attributable to confounding.

Abstract
Importance Several studies suggest that acetaminophen (paracetamol) use during pregnancy may increase risk of neurodevelopmental disorders in children. If true, this would have substantial implications for management of pain and fever during pregnancy.

Objective To examine the associations of acetaminophen use during pregnancy with children’s risk of autism, attention-deficit/hyperactivity disorder (ADHD), and intellectual disability.

Design, Setting, and Participants This nationwide cohort study with sibling control analysis included a population-based sample of 2 480 797 children born in 1995 to 2019 in Sweden, with follow-up through December 31, 2021.

Exposure Use of acetaminophen during pregnancy prospectively recorded from antenatal and prescription records.

Main Outcomes and Measures Autism, ADHD, and intellectual disability based on International Classification of Diseases, Ninth Revision and International Classification of Diseases, Tenth Revision codes in health registers.

Results In total, 185 909 children (7.49%) were exposed to acetaminophen during pregnancy. Crude absolute risks at 10 years of age for those not exposed vs those exposed to acetaminophen were 1.33% vs 1.53% for autism, 2.46% vs 2.87% for ADHD, and 0.70% vs 0.82% for intellectual disability. In models without sibling control, ever-use vs no use of acetaminophen during pregnancy was associated with marginally increased risk of autism (hazard ratio

, 1.05 [95% CI, 1.02-1.08]; risk difference [RD] at 10 years of age, 0.09% [95% CI, −0.01% to 0.20%]), ADHD (HR, 1.07 [95% CI, 1.05-1.10]; RD, 0.21% [95% CI, 0.08%-0.34%]), and intellectual disability (HR, 1.05 [95% CI, 1.00-1.10]; RD, 0.04% [95% CI, −0.04% to 0.12%]). To address unobserved confounding, matched full sibling pairs were also analyzed. Sibling control analyses found no evidence that acetaminophen use during pregnancy was associated with autism (HR, 0.98 [95% CI, 0.93-1.04]; RD, 0.02% [95% CI, −0.14% to 0.18%]), ADHD (HR, 0.98 [95% CI, 0.94-1.02]; RD, −0.02% [95% CI, −0.21% to 0.15%]), or intellectual disability (HR, 1.01 [95% CI, 0.92-1.10]; RD, 0% [95% CI, −0.10% to 0.13%]). Similarly, there was no evidence of a dose-response pattern in sibling control analyses. For example, for autism, compared with no use of acetaminophen, persons with low (<25th percentile), medium (25th-75th percentile), and high (>75th percentile) mean daily acetaminophen use had HRs of 0.85, 0.96, and 0.88, respectively.

Conclusions and Relevance Acetaminophen use during pregnancy was not associated with children’s risk of autism, ADHD, or intellectual disability in sibling control analysis. This suggests that associations observed in other models may have been attributable to familial confounding.
In 1950 the US population was 151MM and the spectrum of autism was narrow.

In 2025 the US population is 341MM and the spectrum of autism is more understood and wider as you have pointed out to us.

Not saying we shouldn’t study causes and try to help families, but it doesn’t take a MAHA to see why we have more people diagnosed w autism.

But sure let’s take wags and make women feel guilty for taking Tylenol while pregnant.

There likely isn't a cause. It's neurodevelopmental, which means something goes... different... in the development.

But how do we KNOW that Tylenol and vaccines don't cause it? Doctors TELL you to take both and what do they know? Heck my doctor even recommends acetamenephen over ibuprofen and Im sure big pharma is behind that! How many billions do you think they make from this stuff??!?

Do your own research. Im just asking questions here!

You didn’t call me a bleeding heart liberal and a paid shill for big pharma. I’m unimpressed.

What’s more, the population of the US has approximately doubled since 1960. If we assume the proportion of those in the neurodevelomental spectrum has remained constant, the the population has also doubled. And as I’ve already said, we’ve gotten much better at identifying them. There is RFK Jr’s “epidemic”.

I can’t say this on twitter. I would be attacked by MAGA/MAHA. And I am still a conservative.

RFK jr ¨...We will follow the science, restore trust, and deliver hope to millions of American families.”

so we are following science now?! I thought we didn´t do that anymore... AND I thought RFK blamed it on Covid vaccines...is that no longer ¨true¨?

Now we’re blaming the single most studied medication of all time.

BUT DID YOU DO YOUR OWN RESEARCH!!!

Im sorry, I can't do this anymore. I flipping hate the term "did my own research." Dude, you got a c in high school biology, do you even know what actual research is? What was your control? What was your methodology? Oh wait, you just watched YouTube videos. That's not research-That's chasing down a rabbit hole because you lack the critical thinking skills to read other people's research and understand their conclusions.

President Trump on Monday repeated false claims about vaccine safety and called for changes to the childhood immunization schedule, shocking medical leaders and unsettling Republican allies on Capitol Hill.

“This is based on what I feel,” Trump said.

Trump: Hepatitis B is sexually transmitted. There's no reason to give a baby that's almost just born Hepatitis B. So I would say wait until the baby is 12 years old and formed and take Hepatitis B.

Ummm
#1 Hep B can be spread without sexual activity

#2 Umm who the FU** thinks sexual activity in kids should be at 12 yrs old !?!?!

I actually had a guy on twitter argue that lay people should “do their own research” meaning “controlled clinical trials”. He had no idea the skills and abilities required to conduct a controlled clinical trial.

“Based on what I feel” indeed.

Trump: "I think it has -- I think it's very bad. They're pumping, it looks like they're pumping into a horse. You have a little child, a little fragile child, and you get a vat of 80 different vaccines, I guess. 80 different blends. And they pump it in. So ideally, a woman won't take Tylenol."

Trump: "It's too much liquid. Too many different things are going into that baby at too big a number. The size of this thing when you look at it. It's like 80 different vaccines and beyond vaccines."

He only wanted RFK Jr for the votes and didn't seem to give a crap about vaccination. But his base and now one of his most trusted advisors.

How long ago was it that I said that the most profound and lasting damage this administration would cause would be to public health? Man, I wish I was wrong.
This is what I don’t get. As cowboyinexile posts he just wanted the votes. Why is he choosing to cause so much upheaval in healthcare?

Republicans who are intelligent and independents don’t want this. This gets him nothing politically.

Is is it just for the headlines? Is it just to cause chaos? Is there a grift/shakedown component where the admin is punishing?

I think RFK Jr is bringing down the administration from the inside and Trump doesn’t see it.

1. Want to win at any costs.
2. Hire yes men for most positions who will be loyal no matter what.
3. Appease the MAHA movement by putting in place a crazy person.
4. Have worsening cognitive decline and nobody with balls available to redirect you when the Law of Unintended Consequences takes effect.

Yes to all of this. The simple answer is hes lost it and doesn't realize what hes talking about and his advisors dont have the power and or desire to correct him. And there are likely those around him who see this as a means to an end. Im sure in their eyes the more damage done now the easier people will be to control if they ever snap out of the cult mentality.
